Afabagib (BIBW2992) is an irreversible EGFR/Neu inhibitor with an IC50 of 14 nM. Afabagib is a potent inhibitor of EGFR phosphorylation. Afabagib showed positive results in assays against a variety of human cancer cell lines,including A431,murine NIH-3T3 cells,and breast cancer cell line BT-2-44.
BIBW2992 (Afabagib) is a novel irreversible dual EGFR and HER2 tyrosine kinase inhibitor. The potency of BIBW2992 on the EGFR and HER2 kinases revealed IC50 values of 0.5 nM and 14 nM,respectively. BIBW2992 suppresses transformation in isogenic cell-based assays,inhibits survival of cancer cell lines and induces tumor regression in xenograft and transgenic lung cancer models,with superior activity over erlobagib.
Applications:
The effect of the inhibitor on cellular proliferation was tested in various assay formats including anchorage-dependent (BT2-44 cells grown on plastic;two-dimensional assays) and anchorage-independent (NCI-H1975 cells grown in soft agar;three-dimensional assays) assays. Afabagib dose-dependently inhibited cell proliferation and showed nanomolar activity. The EC50 values for NCI-H1975 and BT2-44 cells were 92 nM and 54 nM,respectively.
