The isolation of human umbilical vein endothelial cells in AcceGen is from umbilical vein vascular wall by collagenase digestion and culture. Each vial contains ≥5×10^5 cells with high quality. Cells are cryopreserved at the end of primary culture in cell culture media kits with 10% DMSO and delivered frozen. HUVECs are guaranteed a further expansion of the population by 15 times when cultured with AcceGen recommended culture medium and culture protocols. HUVECs are tested for specific antibodies against vWF/Factor VIII, CD31 and uptake of Dil-Ac-LDL by immunofluorescent approach. All cells lots are detected negative for the presence of HIV-1, HBV, HCV, mycoplasma, bacteria, yeast and fungi. AcceGen recommends handling HUVECs with appropriate biosafety procedures due to its human origin. If not using HUVECs immediately, transfer cells to liquid nitrogen (-180℃) until ready for use. Human Umbilical Vein Endothelial Cells (HUVECs) are cells from the vein of umbilical cord. They have cobblestone morphology with large dark nuclei when observed under light microscope. HUVEC cells can be obtained from discorded umbilical cord after childbirth by enzymatic digestion of the lumen. They express many important endothelial signal molecules, for example von Willebrand factor (vWF), which is associated with vascular homeostasis regulation, blood platelets mediation and blood vessel wall tone.
Application
Human umbilical vein endothelial cells culture can be used as a laboratory model for studying vasculature and angiogenesis. They are also good cell source to build scaffolds and facilitate vascularisation in tissue engineering.
